Determination of bile salt
Bile:- it is a yellowish color fluid that is secreted by the liver and stored in the gall bladder.
Method:- Hay’s Method
Principle: –
Bile salt when present on the surface tension of urine when sulfur powder is added to the surface of urine sulphur particles sink to the bottom of the test tube.
In the case of normal urine, sulfur particles float on the surface of urine.
Requirement: –
- Test tube
- Test tube rack
- Sulfur powder
- Urine Simple
Procedure: –
- Take a clean and dry test tube.
- Take about 5 to 10 ml of urine in a test tube.
- Add a Pinch of dry sulfur powder surface of the urine.
- Then observe the sulphur particle.
Observation: –
Sulfur particles sink to bottom bile salt –
present.
sulfur powder floats on the surface bile salt – is absent.
Clinical significance: –
- Jaundice
- Anemia
