Method :- Rothera test
Principle:-
Nitroprusside used for this test reacts with both acetone and acetoacetic acid in the presence of alkali to produce a purpled Coloured..
–Requirement:-
- Test tube
- pipette
- Test babe back.
- Liquor ammonia solution
- -pasteur pipettes-
- Rothera powder mixture
(A).sodium nitroposide-0.75g
(A).ammonium Sulfate- 20 gm
Procedure:-
1) First of all take 5 ml of urine with the help of pasture pipettes in a clean and dry tube
2). Add one gram of rothera reagent powder and mix well properly.
3). After mixing add one or two ml of concentrated ammonium hydroxide.
4). Observe pink purple ring at the interface.
Observation:-
- No appearance of pink purple ring then keton bodies are absent.
- The appearance of a pink-purple ring and then a keton body are present.
Clinical significance :-
-Diabetes mellitus
-Acute infection
-Anorexia
-fasting
-prolong vomiting
-fever
