Determination of Urobilinogen


Principle: –
Urobilinogen reacts with para dimethyl amino benzaldehyde in an acidic medium (in Ehrlich reagent) to form pink colored compound appearance of pink color indicate the presence of urobilinogen in Urine.
Method: – Ehrlich’s method 
Requirement: –
Ehrlich reagent
Barium chloride
Filter paper
Funnel
Test tube 
Rack
Centrifuge 
Pasture pipette
Composition of Ehrlich reagent:-
Para dimethyl amino aldehyde – 2ml
Hydrochloric acid -20ml
Distilled water -30ml/day
Procedure:-
Take 5ml urine with the help of pasture pipettes in the clean and dry test tube.
Add 2.5 ml of 10% Barium Chloride.
Mix well.
Filter the solution in another test tube with the help of filter paper what men filter paper.
Filter solution in 0.5 ml of Ehrlich reagent.
Mix well and wait 3 to 5 minutes.
Result:-(Observation) 
No color change  – negative
Development of pink color – urobilinogen present Normal.0
Development of cherry red color – urobilinogen  Increases

Clinical significance: –
Hemolytic Jaundice 
Hepatic Jaundice
