PACK CELL VOLUME (PCV) Or HAEMATOCRIT VOLUME ( HCT)

Pack cell volume :—- it is a parameter of cbc test.

Definition:– when anticoagulant blood ( double oxalate )is centrifused at standard speed 3000 rpm . And heavy part is settled down in bottom . Column of red cell settle is known as pack cell volume or haematocrit volume 
METHOD :– there are two methods of determination of pcv
1. Macro-haematocrit volume
2.micro-haematocrit volume
SPECIMEN:–2ml of venous blood
PRINCIPLE:–when EDTA blood centrifused then heavy weight substance that is RBC is packed on the bottom of the tube by the centrifugal force .
REQUIREMENTS:–
                   –  wintrobes tube 
                   centrifused machine
                   EDTA vial 
                   timer 
                    cotton swab
                    syringe 💉
                   tourniquet
TESTING PROCEDURE :–
By wintrobes tube method or macro-haematocrit method 

:- collect 2 ml of venous blood in a double oxalate or EDTA vial.

:mixed well properly.

:- mixed blood is fill in the wintrobes tube or PCV tube by the help of pasture pipette on 100 mark.

:- then centrifused the tube at 300 RPM for 20 to 30 minute.
:- after 30 minute stop the centrifused machine and note the level of pcv by the help of marking . 
  
NORMAL RANGE:— 
    IN MALE :- 40 to 52 %
    In female:- 35 to 48 %
    In infant :- 52 to 60 %
