Introduction
The normal white Cell Count 2000 to 11000/cub of blood and these cells can be counted by the help of Haemocytometer.
principle
The whole blood is use for total white Cell Count with dilution Fluid and it is diluted 20 times and placed in haemocytometer then total white blood cell counted under proper spacified area with the help of microscope. 
-Composition of diluting fluid 
1.glacial acetic acid kha 3ml
2.Aqueous mythelene blue  8 to 10 drop
3.distilled water 100 ml 
Requirement
– WBC diluting fluid
– WBC pipette
– Haemocytometer
-microscope
-Blood sample
-Cotton
-Torniquate
-Syringe
-cotton swab
-neubar counting chamber.
-cover slip
-micropipette.

-At 0.5 mark of WBC Pipette or 20 micro liter (0.02ml)of whole blood .
-At 11 mark of WBC diluting Fluid in WBC pipette (0.38 ml)./380 microlitre  
Method
manual
1.draw the Blood up to 0.5 marks in wBC Pipette.
2.- Care Fully clean access blood out side the pipette help of Cotton or tissue paper.
3.-Take the WBC diluting fluid up to 11 mark.
4.Blood and and diluting Fluid mixed well wait 3 to 5 minute.
5.Take neubar chamber and cover slip placed centrally on neubar chamber.
6.- after waitng 3-5 minute to mixed blood taken in WBC Pipette and then few drop dicarded then charge neubar chamber .
7.- neubar chamber charge by capillary action and wait I to 2 minute.
8.After waiting meubar chamber located on mechanical stage of microscope
9.First focus the WBC area with the help of Low power (10x) and finally turn the Objective lense high power (40x) for Counting purpose
10. 4 square areas of WBC are seen.
11.Each square contain 16 smaller squar area.
I squar =16
4 square 4X16 – 64  total Counting square.
– Cell are Identified blue colour with light dot in Centre the square 
Normal Range 4000 to 11000 cub/mm

precaustion
-alway dryy and clea pipette are used
-such the blood exact 0.5 and 11 mark counting chamber should be dry and clean
-give the result care Fully.
