Introduction
– platelet are also known as thrombocyte platelet are small colourless non nucleated cell.
method – manual method.
chemical uses :- Rees euker Fluid or platelet diluting fluid
Composition of Rees Euker fluid
1.Trisodium citrate 3.8 gm
2.Commercial fermaldehyde – 0.2 ml
3.Brilliant cresyl Blue 0.1g (It stain platelet.)
4.distilled water. 100ml.
Specimen
-0.5 mark of blood and 20 micro litre in RBC pipette
– RBC diluting fluid or Rees Euker fluid of 101 mark in RBC pepette.

3.neubar Counting Chamber
4.Rees Euker fluid .

PROCEDURE 
Testing procedure
-draw the blood up to 0.5 mark in RBC pepette.
-CareFully clean the RBC pIpette. outside the pipette extra blood cleaned by tissue power.
– Draw the platelet diluting Fluid up to 101 mark in RBC pipette
– Then mixed properly in horizontal position. and wait for 3-5 minute.
– Cover slip is placed on the centrally neubar Counting Chamber.
-after 3-5 minute waiting of diluted blood discarded few drip .
– A drop of diluted blood is obtain at tip of the pipette.
– drop touch the Junction of coverslip and neubar chamber.
– Charge neubar Counting chamber by Capiliary action wait 1-2 minute.
– Charge neubar counting chamber placed on mechanical stage of microscope. 
– First of all neubar Counting chamber focus by low power (10x) then finally turned the objective lense at high power (40x) for counting purpose.
-There are 25 square area in platelet Counting Chamber. 
– we are Count 5 square of Platelet chamber 4 corner and one center square.
– Cell are identified in dot type form.
Normal Range
in male – 1.5 to 4.5 lakh/cumm
In female-  same
Calculation  
= No.of cell count x 2000
Precaution
-always use cleaning and dry RBC pipette
– suck the blod  exact 5 mark and diluting fluid up to 101 mari
-Neubar chamber clean and dry
– give the result carefully.
Clinical significance 
Increases – Thrombocytosis.
– polycythemia 
-Acute Haemarrhage.
decreases – Thrombocytopenia
                    – chronic imfection
                    – Leukemia
                    – Hypo plastic
                    – Bone marrow disorder
